抄録: In order to assess the regioselective activation of flavin through hydrogen-bonding, new flavins which can form intramolecular hydrogen-bonding between N(1) and pendent NH3+ or phenolic OH were synthesized. The reactivities of these "N(1)-hydrogen-bonded" flavins were not so different from those of conventional flavins. The failure to demonstrate the hydrogen-bonding effect arises probably from (i) the remaining rotational. freedom of the hydrogen-bonding and (ii) the relatively low pKa of N(1) in reduced flavins which does not require the aid of acid catalysis. We have found, however, that the rate constants for the reaction of N(1)-hydrogen-bonded flavins and an NADH model compound increase with lowering the medium pH. This may be taken as evidence for partial contribution of hydrogen-bonding.
