Abstract: The present paper describes the culture conditions of Bacterium No. 101 for production of hemicellulase. The paper also deals with ammonium sulphate fractionation of the enzyme and the enzyme activity assay. The results obtained are as follows: 1) The bacteria was aerobically grown at 37℃ for 7 days in a medium containing of 0.5% hemicellulose, 0.2% NaNO3, 0.1% K2HPO4 and with or without MgSO4. The addition of MgSO4 to a final concentration of M/50 caused 3-fold increase in the hemicellulase production. 2) The supernatant was fractionated by ammonium sulphate and the enzyme activity was found in the precipitate of the 0.25-0.7saturation. 3) To I ml of a 1.0% solution of corn seed hemicellulose in 0.025 M phosphate buffer, pH 5.7, was added I ml of enzyme and after 5 min incubation at 37℃, the reducing sugar formed was determined as xylose by the Somogyi-Nelson method. The enzyme quantity that produced 10 μg of xylose under the above condition was defined as I unit enzyme activity. / 1.糖化力測定法は,pH5.7,0.025M リン酸緩衝液に,基質濃度を全糖量として1.0%に溶解し,一夜放置後不溶解物を除いた基質溶液1mlに酵素液1mlを加え,37°Cで5分間保った後,生成する還元糖をSomogyi-Nelson法にて測定し,キシロース10μg/mlに相当する還元力を生ずる場合の酵素力を1単位とした。2.酵素を最大に産生せしめるためには,基本培地にM/50硫酸マグネシウムを加え,液量をlOOOml容三角フラスコに300mlとして培養するのが適当である。3.粗酵素のpH安定性5~6の間で,熱安定性は,40℃までであった。4.出発酵素液の硫安分画範囲は0.25-0.7飽和が適当であった。
