Abstract: Methods are described to isolate Japanese encephalitis virus (JEV) from field-caught C. tritaeniorhynchus by inoculation into A. albopictus clone C6/36 cell cultures. The methods have similar or even better efficiency to isolate JEV and Getah (GET) virus compared with the classical method of inoculation into suckling mouse brains (SMB). The new methods are also able to isolate such mutants of JEV and GET or other unknown viruses that are hard to detect by SMB inoculation. Several additional advantages of the new methods over the SMB inoculation are described.
